The senior Molecular Biologist, in the Therapeutic Antibody Laboratory (TAL) in the CTSI, will be responsible for molecular cloning of antibody heavy and light chain variable regions from single antigen-specific B cells and hybridomas to support antibody discovery. The senior Molecular Biologist will also be responsible for the preparation of next generation sequencing samples from mouse and human B cell repertoires. The senior Molecular Biologist will be experts in state of the art techniques for sequencing and cloning of antibodies and proteins in general, with experience in bacterial and mammalian vectors supporting protein production. Will collaborate with scientists within the organization to actively improve therapeutic antibody discovery efforts by developing state of the art molecular biology and sequencing technologies, and their integration with automation and informatics tools. The incumbent may also coordinate activities and experiments with partner laboratories across UCLA and the DGSOM.
Establish efficient work flow for molecular cloning of immunoglobulin genes from various species and sources (e.g. mouse hybridoma, single human and murine B cells)
Establish and optimize a high throughput workflow to study the immunoglobulin repertoire
Analyze traditional and next generation sequencing data
Drive novel antibody discovery technology development
Design and construction of expression vectors for the production of recombinant antigens and antibodies in bacterial and transient and stably transfected mammalian cells
Conduct independent research projects, organize research data and present to lab members, finalizing research outcomes by composing and publishing research articles.
BS or MS (with minimum 8-12 years experience) or PhD (with minimum 4-6 years experience) in immunology, biology or similar field with application to antibody and protein cloning and expression
Strong proven molecular biology skills
Proven expertise in Ig amplicon generation from immune repertoires, hybridomas and single B cells
Working knowledge in one or more NGS sequencing platforms
Knowledge and proven record of developing state of the art antibody discovery platforms
Expertise in antibody V gene sequencing and cloning
