Job Code: 4952 Job Grade: G The Center for Receptor-Ligand Discovery at Stanford University School of Medicine is seeking a highly-motivated Life Science Research Professional 2 to join a biotechnology-centered lab in the Department of Molecular and Cellular Physiology. The successful candidate will perform research functions and activities for defined projects involving molecular biology, cell biology, protein biochemistry and protein engineering to support the maintenance of existing research projects and, additionally, will contribute to both the optimization of current assays and the development of new technologies. The LSRP2 is expected to independently conduct, analyze and present experiments under the support and guidance of postdoctoral fellows and other staff members in the laboratory. S/he will keep meticulously detailed records of experiments and prepare results of his/her research for inclusion in reports, presentations and publications.
The successful candidate will work with a multidisciplinary and dynamic research team on a genome-wide project to study cell surface receptor-ligand interactions as part of a collaborative research initiative entitled the Human Interactome Project. The goal of this project is to create an atlas of all human cell-surface interactions via high-throughput screening of receptors and ligands using existing assays and through the development of new technologies. Interactions at the cell surface play a crucial and widespread role in regulating biological processes and, as such, de-orphanizing receptor-ligand interactions is essential for understanding biological processes and may provide valuable therapeutic targets for drug discovery which have the potential to improve human health in the context of many disease states.
The LSRP2's contributions to this project will principally involve gene construct design and cloning, protein engineering, expression and purification from insect and mammalian cells, Western blotting, in vitro protein-protein interaction assays and cell-based assays. While some limited degree of on-the-job training is acceptable for desired areas, this will be determined on a case-by-case basis and depend on the additional skill set the candidate possesses. Communication skills (both oral and written) will be weighed heavily.
Plan and perform existing experiments in support of ongoing research projects based on knowledge of scientific theory. Interpret and perform analyses of results, often where few guidelines exist. Based on prior results, contribute to development of new research protocols and procedures or adaptation of existing protocols.
Make recommendations on plan design and/or research direction to project manager and principal investigator. Offer suggestions regarding modifications to procedures and protocols in collaboration with graduate students, postdoctoral fellows and staff.
Perform carefully-designed, meticulously-executed and well-documented optimization experiments to assist in the development of novel assays to detect receptor-ligand interactions, establish new lab protocols and recommend appropriate modifications to existing ones.
Molecular biology techniques including PCR, cloning, gel electrophoresis, plasmid purification, sequence analysis, construction of expression vectors for production of recombinant proteins to be used downstream in in vitro and in vivo assays.
Tissue culture including transformation, transfection and infection of bacterial, insect and mammalian cells for virus amplification/production and protein expression.
Purification (affinity tag and size exclusion chromatography) of recombinant proteins expressed in bacterial, insect and mammalian systems to study receptor-ligand interactions.
Protein biochemistry techniques (e.g., Western blotting) to characterize recombinant proteins, in vitro protein-protein interaction assays (e.g., ELISA, SPR, co-IP) and cell-based binding assays using flow cytometry, FACS and fluorescence microscopy analyses.
Contribute to bioinformatics analyses of human cell surface and secreted proteins.
Review literature on an ongoing basis to remain current with new procedures and related research.
Maintain meticulously detailed records and documentation of experiments, observations, data and analyses.
Contribute substantively to the preparation of papers for publication, especially in the results section. May contribute to publication of findings and present ongoing work and findings to colleagues at academic conferences. Participate in the preparation of other written documents, including procedures, presentations, proposals and progress reports.
Participate in multidisciplinary teams across different faculties or schools.
Work with health and safety department to ensure lab complies with regulations and all required trainings.
Oversee supply and equipment budget under supervision of principal investigator or business manager.
Supervise (either formally or informally) staff or students as needed, including oversight and instruction on techniques, as well as consultation on project work. Serve as technical resource for other research staff.
Perform general lab maintenance as needed; maintain lab stock, manage chemical inventory and safety records, and provide general lab support as needed. May assist with purchasing laboratory equipment.
