The California Institute for Quantitative Biosciences (QB3) is one of four Governor Gray Davis Institutes for Science and Innovation established in 2000 to ensure the future of the California economy by promoting research and innovation. QB3 is a cooperative effort between the state of California, private industry, venture capital, and the University of California campuses at Berkeley, San Francisco, and Santa Cruz.
QB3 harnesses the quantitative sciences of physics and engineering to unify our understanding of biological systems at all levels of complexity, from atoms and molecules to cells, tissues, and entire living organisms. The Staff Research Associate will, under supervision, contribute to the production of novel reagents enabling genome editing of specific cell types, with a focus on enabling accessible therapeutic use of this technology. This position will involve molecular cloning, biochemical techniques, chemical biology, and analytical processes dedicated to the production and characterization of molecules enabling targeted therapeutic genome editing. These molecules include monoclonal antibodies or fragments thereof, engineered CRISPR guide RNAs, and RNA-binding proteins conjugated with functional molecules. The SRA will generate and characterize these molecules. As needed, the SRA may also contribute to the generation of engineered cell lines via viral transfection.
Generate monoclonal antibodies and fragments thereof. This work may involve maintenance of multiple cell types and molecular cloning to generate the necessary genetic constructs. Resulting molecules will be isolated and assessed for the purity and desired identity.
Generate engineered CRISPR guide RNAs. This work involves molecular cloning to generate the desired template construct, molecular biology to produce large quantities of said template, and an in vitro transcription reaction followed by gel-based purification. Resulting molecules will be assessed for quality using a denaturing gel and/or mass spectrometry.
Generate conjugated molecules enabling genome editing. This work involves using custom-generated RNA-binding proteins as a hub for localizing peptides to the engineered RNA molecules described above. To do this, conjugation reactions will be performed, followed by purification and analysis of purity and identity. Because each peptide has distinct chemical properties, conjugation and purification steps often require optimization, making this work non-routine and necessitating a capable scientist to complete the work efficiently.
Use lab computers and software to record and compile data; prepare results for presentation.
B.S. in a relevant field (degree must be completed before start date; not required to apply)
1 year of part-time (10 hours per week) or full-time experience in a life-sciences laboratory setting
Basic life-sciences laboratory skills:
Using a pipettor, preparing solutions, diluting reagents, following an established protocol
Working knowledge of MS Office suite (Word, Excel, Powerpoint)
Detail-oriented and well-organized in terms of record-keeping, planning ahead, meeting deadlines
Experience with DNA amplification, purification, and analysis (PCR, Sanger sequencing, etc.)
Experience with RNA/protein biochemistry
Experience with tissue culture, especially mammalian tissue culture
Experience with molecular cloning
Experience with antibodies or fragments thereof
